Sawada Tsunayoshi or Tsuna is the main character of the series - he's the successor to the 9th boss of the strongest mafia family in the world: The Vongola. As Tsuna starts out in the series, he doesn't actually seem to be much of anything. He doesn't make good grades, isn't well-liked by the anyone, has a major crush on Sasagawa Kyoko (popular girl in school) -- just in general, a loser. Heck, even his own mother doesn't see much in him. Even so, the 9th boss of the Vongola chose Tsuna to be the successor, and Tsuna's fate is inevitable. He just needs proper training from Home tutor Reborn, right? Tsuna stumbles a lot and doesn't really have any idea of what he's doing, what he's up against, he's just worried for his friends. That alone - protection of friendship and the care of others - is what makes him the Vongola 10th. He's a pretty weak kid, but in terms of hurting friends and others...? He's not a force to be reckoned with.
That same driving force, the one that cares so deeply for those around him, is the force that causes Mukuro's defeat in the first season. Being that Mukuro is Tsuna's challenge in the first season, Mukuro doesn't actually see much in Tsuna being a major threat. The real threat kind of lies with his subordinates like Gokudera, Yamamoto and Bianchi. Once Tsuna makes it to Mukuro's headquarters, Mukuro begins to be very cautious of the young Vongola. During Tsuna's battle with Mukuro, Mukuro constantly mocks him and teases him, using illusions and nightmares to weaken Tsuna's will and force him into following his order. Tsuna, not knowing what to do, is in panic for most of that battle until Leon spits out Tsuna's new weapon.
After Tsuna proves himself to be stronger than Mukuro, everything about this relationship changes. From Mukuro thinking that Tsuna is a weak kid with no merits and nothing will become of Vongola 10th to warning others about Tsuna's strength can be a huge jump for Mukuro. I don't think Mukuro is the type to admit others are far superior to his power, anyway. For Mukuro, he believes that Tsuna has great strength and is, in fact, fit for leading the Vongola. It still doesn't really change the fact that Mukuro wants to take over his body (which can almost be seen as an empty threat? it depends).
Though, in light of all this... Tsuna never feels animosity for his enemies. Not even for Mukuro, who planned to kill everyone in his own family, the Vongola. After Mukuro was arrested again, Tsuna had wondered if Mukuro and his two subordinates were alright - and hoped that the punishment didn't kill them. Luckily, it didn't, which relieved Tsuna. In a sense, Tsuna has an inner desire to make good with Mukuro and doesn't want to see Mukuro as an "enemy" any longer.
